What the company does
Corcept Therapeutics discovers and develops medications for endocrinologic, oncologic, metabolic and neurologic disorders by modulating the effects of the hormone cortisol. Since 2012 it has marketed Korlym in the United States for patients with hypercortisolism, also known as Cushing's syndrome, and in June 2024 it made an authorized generic version of Korlym available for the same indication.
The science
Korlym's active ingredient, mifepristone, reduces the binding of excess cortisol to the glucocorticoid receptor (GR). Mifepristone also binds to the progesterone receptor (PR), which terminates pregnancy and causes other adverse effects, including endometrial thickening and vaginal bleeding. The company has discovered more than 1,000 proprietary cortisol modulators that bind to the GR but have no affinity for the PR. They fall into four structurally distinct series and include relacorilant, nenocorilant, miricorilant and dazucorilant.
