- Investigator-assessed PFS also improved. Investigator-assessed median PFS was 6.7 months with the combination versus 2.7 months in the ICT arm, with hazard ratio 0.36. Management described the safety profile as generally well-tolerated.
- Overall survival data still immature. Overall survival data were immature at the January 23, 2026 cutoff. Management noted an early trend in OS improvement with the combination.
- Full primary data shown at ASCO. Complete primary-analysis data from 313 patients were presented as a late-breaking oral at the 2026 ASCO meeting in Chicago.
- OptimUM-01 enrollment completed at about 100. Enrollment in the single-arm Phase 2 OptimUM-01 trial is complete at approximately 100 HLA*A2:01-positive patients. Data from about 85 efficacy-evaluable patients are planned for ESMO.

What the company does
IDEAYA Biosciences is a precision medicine oncology company that discovers and develops cancer therapies. It works on small-molecule drugs and antibody-drug conjugates (ADCs) for solid tumors defined by their genetics. The company describes its clinical pipeline as nine potential first-in-class product candidates across four clinical focus areas.
The four focus areas
The first area is darovasertib for uveal melanoma. The second is ADC and DNA Damage Response (DDR) combinations.
The third area is the MTAP pathway, which targets tumors with MTAP gene deletions. The filing says these deletions occur in approximately 15% of all solid tumors. The programs here are IDE397, a MAT2A inhibitor, and IDE892, a PRMT5 inhibitor.
Darovasertib and uveal melanoma
Darovasertib is an oral small molecule inhibitor of protein kinase C (PKC) and the company's most advanced program. Approximately 95% of uveal melanoma patients carry activating mutations in GNAQ or GNA11. In metastatic disease, the company is testing darovasertib with Pfizer's crizotinib in OptimUM-02, a Phase 2/3 trial that has enrolled 437 patients. In primary disease, OptimUM-10 is a randomized Phase 3 neoadjuvant trial planned for approximately 450 patients.
The FDA has granted darovasertib Orphan Drug designation in uveal melanoma, Fast Track designation for the crizotinib combination in metastatic disease, and Breakthrough Therapy designation as a neoadjuvant treatment for primary disease. The company keeps all rights to darovasertib in the United States.
Partners and licences
Darovasertib is licensed from Novartis, with worldwide rights. In August 2025 the company granted Servier an exclusive licence to develop and commercialize it everywhere except the United States. Servier is responsible for regulatory and commercial work outside the United States, and the two companies share development costs. The filing lists royalties on those sales ranging from mid-teens to low-twenties percentages.
Pfizer supplies crizotinib for the darovasertib trials. Gilead supplies Trodelvy for trials combining it with IDE397 in urothelial cancer and non-small cell lung cancer, and each company keeps its own commercial rights. IDE161 is licensed from Cancer Research UK and the University of Manchester. IDE849 is licensed from Hengrui Pharma for all territories outside Greater China.
The Pol Theta (IDE705) and WRN (IDE275) programs transfer back to the company.
How it finds drugs
The company's discovery work combines small-molecule chemistry, structural biology and bioinformatics with biomarker work. Its INQUIRE chemical library covers over 200,000 chemical compounds. It also uses a machine-learning engine called HARMONY for molecular design.
The DECIPHER dual CRISPR library, built with the University of California, San Diego, allows evaluation of approximately 50,000 independent gene knockout combinations. A collaboration with the Broad Institute searches for synthetic lethality targets in ovarian cancer, and the company is a member of the Broad DepMap consortium. For patient selection, it has relationships with Foundation Medicine, Guardant Health and Tempus.
Manufacturing and selling
Contract manufacturers make the active ingredients and finished drugs for its small molecules, including STA Pharmaceutical, Yuhan Corporation, Patheon and Pharmaron. Biocytogen and Hengrui Pharma supply the in-licensed ADCs for development.
The company says it intends to become a fully integrated commercial stage biopharmaceutical company.
Competition and protection
The filing describes the industry as very competitive, with major pharmaceutical, specialty pharmaceutical and biotechnology companies in the field. In uveal melanoma, it names Immunocore's Kimmtrak as a commercialized therapy. For IDE849, it names Amgen's Imdelltra, a DLL3-directed therapy that received FDA accelerated approval.
As of 30 January 2026, the company owns or exclusively licenses patents and applications in approximately 71 distinct patent families. These include approximately 21 issued United States patents and 65 issued foreign patents.
Its people
The team is led by Chief Executive Officer Yujiro S Hata and includes cancer biologists, drug discovery chemists, translational biologists and drug development professionals. A scientific advisory board also guides the company.
